氣道平滑肌是維持氣道張力的根本。它存在于氣管、支氣管樹直至終末支氣管。其特異的超微結構特點和調節機制是實現氣管正常生理功能的基礎。有證據表明氣管平滑肌在肺的發育過程以及諸如**病、慢性支氣管炎和肺氣腫等疾病狀態下經歷著顯著的表型變化。另外,氣道平滑肌通過分泌細胞因子和趨化因子等促炎介質,發揮免疫調節功能。培養的氣管平滑肌細胞能表達三種P21活化的激酶(PAK)的同型異構體和PAK1,并通過P38 mapk信號途徑參與氣管平滑肌細胞的遷移。氣道平滑肌細胞增殖已成為人們關注的焦點,因為在**病人中,此細胞是氣道壁重塑反應中數量上的重要組分,并被認為是新型抗**病藥物的作用目標。
人氣管平滑肌細胞提取于人氣管組織,二代凍存。每管含有細胞數>5×105 cells/ml,此細胞通過對alpha-SMA和Desmin免疫熒光染色驗證,經測試不含有HIV-1、HBV、HCV、支原體、細菌、酵母和真菌。細胞可以達到15倍增。? ?
