心肌細(xì)胞是機(jī)體*具有活力的細(xì)胞。它們是高度分化的高氧含量的細(xì)胞,擁有大量的線粒體。心肌細(xì)胞占有心臟重量的75%,但僅為心臟細(xì)胞數(shù)量的1/3。高度分化的心肌細(xì)胞幾乎沒有增殖能力,它們在a-腎上腺素的刺激下通過Ras/MEK途徑發(fā)生肥大生長。所有的心肌細(xì)胞都能自律性的將膜去極化和復(fù)極化。所有的心肌細(xì)胞收縮都是肌源性的,不受神經(jīng)系統(tǒng)刺激。心肌細(xì)胞具有復(fù)雜的信號網(wǎng)絡(luò),從而調(diào)節(jié)其在心臟節(jié)律性的泵出功能中的作用。心力衰竭時,心肌細(xì)胞的肥大和凋亡導(dǎo)致心肌收縮功能的下降。更好地了解心臟信號系統(tǒng)有助于揭示心肌細(xì)胞死亡的細(xì)胞機(jī)制。
Sciencell研究實(shí)驗(yàn)室的HCM是從人心臟分離得到的,原代凍存,冰凍運(yùn)輸。每管細(xì)胞密度超過1×10^6/ml。細(xì)胞特性經(jīng)肌球蛋白(myosin)免疫熒光鑒定。本細(xì)胞經(jīng)檢測不含HIV-1、HBV、HCV、支原體、細(xì)菌、酵母菌和真菌。如采用ScienCell?實(shí)驗(yàn)室提供的培養(yǎng)條件,可保證此細(xì)胞繼續(xù)的培養(yǎng)。HCM因?yàn)椴荒茉鲋常什荒軌驍U(kuò)大或長期培養(yǎng)。?
